中文摘要: 检测循环肿瘤细胞对癌症患者的早期诊断及疗效评估等具有重要意义。本项目拟以非小细胞肺癌循环肿瘤细胞为研究对象,采用肿瘤细胞表面高表达表皮生长因子受体的抗体和适体作为分子识别物质,结合生物及电化学双功能化石墨烯纳米复合材料,用于构建肿瘤细胞电化学敏感仿生传感界面,建立捕获肿瘤细胞新方法,研制灵敏、准确、快速的循环肿瘤细胞电化学生物传感器。探索石墨烯的功能化方法,建立能保持抗体和适体对肿瘤细胞亲和力的固定化方法,利用电极表面固定化的识别物质高效捕获肿瘤细胞;深入研究在传感界面上捕获肿瘤细胞新方法及非特异性吸附干扰等问题。拟选用白细胞膜抑制正常体细胞在传感界面上的非特异性吸附。本项目力求建立检测循环肿瘤细胞新方法,并将其用于非小细胞肺癌的早期临床诊断和肿瘤细胞与抗肿瘤药物相互作用研究。该项目将为循环肿瘤细胞检测的基础和应用研究提供器件和分析新方法,将促进肿瘤细胞检测相关生物传感器和生物芯片的研究。

英文摘要: Circulating tumor cells (CTCs) analysis is a promising new diagnostic field for the early diagnosis and estimating the risk of metastatic relapse and metastatic progression in patients with cancer. The project intends to choose non-small cell lung cancer (NSCLC) as research model, and to construct the biomimetic sensing interface which sensitive to tumor cells using antibody and aptamer of the high expressed epidermal growth factor receptor (EGFR) of NSCLC tumor cells as molecular recognition substances and combining with biologically and electrochemically functional graphene nanocomposites; intends to develop several novel electrochemical biosensors and to establish new methods with sensitivity, selectivity and speediness for the detection of NSCLC circulating tumor cells. To exploit the functionalization methods of graphene, to establish the immobilization methods of antibody and aptamer which can be maintained their affinity for tumor cells, and to efficiently capture tumor cells using the immobilized recognition substance on the electrode surface; To lucubrate on the on-line capture methods of CTCs and non-specific adsorption interference of human normal somatic cells on the biomimetic sensing interface, and intends to use the membrane of human leukocyte to suppress non-specific adsorption of human somatic c
