中文摘要: 季铵生物碱是生物碱中的一大类活性组分,具有非常好的药理活性,对其代谢动力学研究对于阐明其作用机制,设计及优选给药方案,促进新药开发和剂型改进及质控等具有重要意义。而开展其代谢动力学研究的关键就是要建立复杂生物样品中微量季铵生物碱的快速、准确、有效的分析方法。氧化石墨烯是一种新型的二维碳纳米材料,表面含有大量含氧活性基团,是非常好的极性化合物的萃取和分离材料,但由于其片状纳米结构,很难直接应用到萃取和色谱分离中。本项目拟利用氧化石墨烯的优异特性,制备氧化石墨烯掺杂的整体SPE小柱对生物样品中微量季铵生物碱进行高效富集,制备氧化石墨烯包覆硅胶的极性色谱分离材料对所提取的季铵生物碱进行有效分离,并与质谱联用进行检测,进而建立复杂生物样品中微量季铵生物碱的萃取、分离、检测三位一体的分析新方法。该项目的顺利实施将进一步拓宽氧化石墨烯的应用领域,为季铵生物碱药代动力学研究提供有力的分析技术支撑。

英文摘要: Quaternary ammonium alkaloids with excellent pharmacological activities, is class of active ingredients of alkaloids. The pharmacokinetic study of quaternary ammonium alkaloids is very important to elucidate mechanism of action, design and optimize dosage regimen, promote the development of new drugs and dosage improvement and control quality etc. The key to carry out the pharmacokinetics is must establishing a fast, accurate and effective analysis method for determination of trace quaternary ammonium alkaloids in complex biological samples. Graphene oxide with a large number of oxygenated active groups on the surface is a new two-dimensional carbon nanomaterial and excellent extraction and separation material for polar compounds. But due to its lamellar nanostructure, it is difficult to directly be used in extraction and chromatographic separation. Using the excellent properties of graphene oxide, this project aims to prepare graphene oxide doped SPE cartridges for high-efficiency extraction of trace quaternary ammonium alkaloids in biological samples, prepare polar chromatographic separation materials with graphene oxide coated silica gel as matrix for good separation of the extracted quaternary ammonium alkaloids, and then combine with mass spectrometry for quantitative detection.
