中文摘要: 肿瘤对人类的健康和生存构成重大威胁,是世界各国面临的最重要的社会问题之一。本项目瞄准多光子成像技术在临床诊断和治疗评估的独特价值以及肿瘤早期诊断和治疗的重大需求,开展肿瘤细胞外基质的多光子成像研究。利用多光子成像技术研究肿瘤进程不同阶段的组织、不同生物学特性的肿瘤标本、活体肿瘤动物模型,探索细胞外基质的内源性多光子信号及显微结构的变化;并结合分子生物学技术揭示肿瘤增殖、侵袭、转移、上皮-间质转化过程中细胞外基质内源性多光子信号的变化;在体监测和评价不同方法治疗过的肿瘤动物模型,分析其细胞外基质的重塑情况以及相应的多光子信号和显微结构的变化。预期成果将实现肿瘤细胞外基质生物形态和生物化学的信息整合分析和可视化,为肿瘤诊断和治疗提供新思路和手段,为多光子成像技术用于临床提供依据和实验数据。

英文摘要: Tumor has threatened people’s health and survival seriously, and it has become one of the most important social problems facing all the countries in the world today. This project is established in the unique value of multiphoton imaging technique in the diagnosis and treatment evaluation of diseases and the significant demand-‘detection and treatment of early tumor’, and focuses on the study of the tumor extracellular matrix based on multiphoton imaging technique. In this project, ex vivo normal, precancerous and cancerous human tissues, tumors with respective biological characteristics, in vivo tumor animal models, and the effects of different therapy options on tumor animal models will be investigated to acquire intrinsic multiphoton signals and microstructural changes. Moreover, molecular biology techniques will be combined to understand the link between intrinsic multiphoton signals and tumor proliferation, invasion, metastasis, epithelial-mesenchymal transition. This project will achieve the integration analysis and visualization of information on the biomorphology and biochemistry of tumor extracellular matrix, and offer the novel tool and the scientific scheme for the diagnosis and treatment evaluation of tumor.
