中文摘要: 独角金内酯(Strigolactones,SLs)是新近发现的一类內源性植物激素,能够抑制植物分枝并调控植物根的生长等生理过程,同时还具有促进寄生植物种子萌发和共生真菌菌丝生长的作用,在调控植物株型和生长发育等方面具有重要的应用前景。目前,SLs的生物合成途径、活性形式、代谢和信号转导等方面还有很多未知问题有待研究,因此能够准确定量分析植物中的SLs 对于阐明这些问题具有非常重要的意义。然而,目前通行的SLs类化合物分析方法存在材料消耗量大、周期长、步骤复杂等问题,特别是在植物样品前处理方面。本研究使用在线固相萃取技术,建立全自动的SLs分离纯化方法,并结合色谱质谱联用技术,建立水稻中高通量、高灵敏度SLs On-line SPE-LC-MS/MS分析方法,以解决其分析方法稳定性差和灵敏度低的问题。通过该方法在植物材料中的应用,为SLs分子作用机理研究提供技术支撑和重要线索。

英文摘要: Strigolactones (SLs) are a novel class of plant hormones, which act as inhibitors of plant branching and regulate other physiological processes such as root growth, as well as induce seed germination of root parasitic weeds and hyphal branching of arbuscular mycorrhizal fungi. All of these indicate that SLs possess important application prospect in regulation of plant growth and architecture. So far, researches on the biosynthesis, potential final bioactive form, transformation of bioactive forms and metabolism and signalling of SLs still remain unambiguous. Quantitative analysis of SLs has great significance to the elucidation of the physiological roles of SLs. However, the current methods are questionable in many aspects, such as large plant tissue consumption, multi-step and labor-intensive. Focusing on the key technologies and procedures of ultra-micro analysis of SLs,this project will explore and optimize an efficient and manageable purification strategy of SLs through automated on-line solid-phase extraction-liquid chromatography-electrospray tandem mass spectrometry(On-line SPE-LC-MS/MS).The aim of this project is to improve the stability and sensitivity of quantative analysis results of SLs in rice.
