中文摘要: 研究发现蓝蕚香茶菜二萜类化学成分具有很强细胞毒作用,无细胞毒的二萜显示很强的抗乙肝病毒活性。新化合物蓝蕚己素有强抗乙肝病毒作用;蓝蕚X素有较强的细胞毒作用。发现二萜结构中无末端双键者抗乙肝病毒作用强,而有末端双键者细胞毒作用强。蓝萼甲素有很强抑制T-CEM细胞作用(IC50(μg/ml)0.049),对HL-60裸鼠模型抑瘤率66%.已获发明专利3项(蓝萼X素制备抗癌药物的应用ZL200910044884.X;蓝萼香茶菜二萜类化合物制备抗癌药物的应用 ZL200910044882.0;蓝蕚香茶菜二萜类化合物制备抗肝炎病毒药物的应用 ZL200910044883.5)。本研究拟用多种现代色谱方法分离活性成分,用多种波谱技术鉴定化合物的结构,评价生物活性,结构活性单元,进行部分结构修饰,研究作用机制和部分构-效关系。为寻找开发具有知识产权的新天然抗癌和抗肝炎病毒的先导化合物提供重要的研究基。

英文摘要: In this research project,we found that diterpenoid compounds from Rabdosia japonica showed strong anti-hepatitis B virus and cytotoxic activities by activity tracking method.The diterpenoids without cytotoxic activity show strong anti-HBV activity.New compound glaucocalyxin F has stronger anti-HBV activity and glaucocalyxin A has very stronger cytotoxic activity(IC50(μg/ml)0.049). We found that the diterpenoids without terminal double bond showed appreciable anti-HBV activity while the diterpenoids with terminal double bond have strong cytotoxic activity.We obtained 3 patents for inventions(Glaucocalyxin X in the preparation of anti-cancer drugs ZL200910044884.X;Diterpenoids from Rabdosia japonica in the preparation of anti-cancer drugsZL200910044882.0;Diterpenoids from Rabdosia japonica in the preparation of anti-hepatitis virus drugs ZL200910044883.5)To isolate new compounds and to search for biologically active compounds from Rabdosia japoncia,we will use a variety of chromatograhic and spectroscopic techniques.Then we will evaluate their biological activities and comfirm activity units of these diterpenoids.Finally,we will modify parts of stuctures of these diterpenoids to study the mechanism of action and structure-activity relationship of them.This study provide an important basis for futher study to find
