中文摘要: 从药用植物中寻找天然抗艾滋病病毒(HIV)活性成分是研发抗艾滋病病毒创新药物的重要途径,为抗病毒药物研究领域的热点。瑞香科瑞香属植物具有抗病毒、抗肿瘤、杀虫等多种生物活性,萜类成分是其主要的药效物质基础。项目组前期从尖瓣瑞香中发现具有极强抗艾滋病病毒的活性萜类成分,显示瑞香属植物可以作为天然抗艾滋病病毒活性成分库进行挖掘,国内瑞香属药用植物资源抗艾滋病病毒活性成分仍值得深入研究。本项目拟在已有研究基础上开展国内三种瑞香属药用植物(橙黄瑞香、丝毛瑞香、白瑞香)中萜类成分的定向分离与鉴定,发现新颖萜类分子,揭示其萜类分子多样性,研究萜类分子抗艾滋病病毒作用机制,获得抗艾滋病病毒活性萜类成分,为三种药用瑞香中天然产物研发自主知识产权抗艾滋病病毒药物以及发现抗艾滋病病毒候选药物或先导化合物奠定科学基础。

英文摘要: Searching natural active constituents for anti-HIV from medicinal plants is the important approach for developing anti-HIV agent targeting for AIDS, and it tends to be one of the hotspot about anti-virus drug study. The medicinal Daphne species were used as medicinal herb for antivirus, anticancer, and insecticide. Terpenoids from those plants are the main active substance. Our research on D. acutiloba previously showed the presence of strongly anti-HIV active constituents, indicating that the natural library of anti-HIV constituents present in Daphne plants. The in-depth study on active constituents from the medicinal Daphne plants growing in China will be performed. This project will carry out the directional isolation and identification of terpenoids from three medicinal Daphne plants (D. aurantiaca, D. holosericea, and D. papyracea) growing in China, investigation on their mechanism of action for anti-HIV, and obtaining novel terpenoids and active constituents for anti-HIV base on previous research, which would lay the groundwork for development of natural anti-HIV agent possessing intellectual property rights from these five Daphne plants and discovery of high efficiency drugs or lead compounds.
