中文摘要： 本研究基于"治未病"理论，创建"针灸预处理诱导脑缺血耐受"假说。通过借鉴脑缺血预适应现象认识针刺、灸法预防缺血性脑卒中的作用机理，提出防治缺血性脑血管病新理念，丰富和发展针灸"治未病"理论。从组织形态学，免疫组织化学，分子生物学角度观察针刺、灸法预适应对全脑缺血大鼠大脑皮层，海马区神经元尼氏体变化，神经元凋亡，Bcl-2，c-fos蛋白的表达和基因mRNA表达的影响，评价针刺、灸法效应，探索防治缺血性脑血管病的新途径，为临床针灸防治缺血性脑血管疾病提供理论依据。 研究结论：①#38024;灸预处理可以延缓或者阻断缺血性卒中的发展进程，增加神经元的存活率；②#38024;灸预处理可以抑制全脑缺血大鼠大脑皮层、海马CA1区神经元凋亡。③#38024;灸预处理可通过上调细胞凋亡基因Bcl-2mRNA和蛋白表达而抑制缺血脑组织神经细胞凋亡。④#38024;灸预处理的预防性脑保护作用可通过促进HSP70mRNA、HSP70蛋白表达高峰提前实现。⑤#38024;灸预处理可通过上调CA1区c-fosmRNA表达而减轻严重缺血后细胞凋亡，促成脑缺血耐受的产生。⑥#38024;灸预处理抑制对脑缺血再灌注大鼠iNOS和nNOS活性，诱导脑缺血耐受。

英文摘要： This study is based on the the theory of "Preventive Treatment of Disease" , establishing a hypothesis of cerebral ischemic tolerance induced by acupuncture and moxibustion pretreatment. By using the successful experience of cerebral ischemic preconditioning ,developing the mechanism of acupuncture and moxibustion pretreatment cerebral ischemic, putting forward the new concept of pretreatment cerebral ischemic, enriching and developing the Theory of "Preventive Treatment of Disease" by the means of acupuncture and moxibustion.Application of HE staining to observe change of histomorphology，electron microscope to observe ultrastructure，TUNEL method to observe apoptosis state，immunohistochemistry and in Situ Hybridization technique to detect Bel-2,c-fos, HSP mRNA and protein expression，evaluating effects of acupuncture and moxibustion, exploring a new approach to pretreatment cerebral ischemic. Conclusions:①cupuncture and moxibustion preconditioning can delay abolish the further advance of ischemic stroke and increase the tolerance of ischemic brain tissue about a series of metabolic disturbances，increase the survival rate of neure，improve the outlook.②cupuncture and moxibustion preconditioning can prevent cell apoptosis in cerebral cortex and hippocampal CA1 section.③cupuncture and moxibustion preconditioning can prevent cell apoptosis in ischemic brain tissue by inereasing Bel-2mRNA and proteinexpression.④he protective mechanism of acupuncture and moxibustion preconditioning may be carry out by early expression of HSP70mRNA and Protein.⑤cupuncture and moxibustion preconditioning can prevent cell apoptosis in ischemic brain tissue by inereasing c-fos mRNA and protein expression.in hippocampal CA1 section. ⑥he ability of acupuncture and moxibustion preconditioning inhibiting the activity of NOS is the same as that of ischemia-reperfusion ,suggesting that moxibustion preconditioning is an effective means for inducing cerebral ischemic tolerance.
