中文摘要: 神经康复是针灸的优势所在。我们前期研究已初步证实,促神经元可塑性是针灸实现神经康复的重要基础,而正常脑能量代谢是神经元发挥可塑性功能的关键,但其具体机制尚不明了。最新研究显示,星形胶质细胞(AS)与神经元进行能量交互,为可塑性的发挥提供能量底物乳酸。预研究显示,针刺能影响机体乳酸水平,那么针灸是否通过影响AS-神经元的乳酸交互促进可塑性呢?其机制是什么?本课题基于前期研究,在中医针灸理论指导下,以脑老化SAMP8小鼠模型为研究对象,运用神经行为学、神经电生理学及分子生物学等研究技术,从乳酸生成、转运、利用这一关键环节入手,结合阻断手段,围绕"AS糖原分解-乳酸生成-MCT4转运胞外-MCT2转入神经元胞内-乳酸利用,ATP生成-神经元可塑性-LTP增强和学习记忆能改善"链条效应进行系统研究,为深入揭示针灸对神经元可塑性的影响机制提供实验依据,为研究针灸的神经康复作用提供新的切入点。

英文摘要: Neurological rehabilitation is one of the advantages of acupuncture. Our previous study confirmed that promoting neuronal plasticity is an important foundation for neurological rehabilitation by acupuncture, and the normal brain energy metabolism of neurons play a key role in neural function, but the mechanism is unclear. The latest studies have shown that the interaction with lactate between astrocytes (AS) and neuronal enhanced synaptic plasticity.Our research result told that acupuncture can affect the body's lactate levels.So,we think that whether acupuncture promote neural plasticity by affecting the lactate shuttle between AS and neuron ? What is the mechanism? For this research,which is based on preliminary studies,and under the guidance of the theory of Chinese medicine and acupuncture ,we will employ many technologies,including neural behavior, neural electrophysiology,immunohistochemistry,insitu hybridization,western blot, real- time PCR and so on ,to detect the key link of lactate production, transfer and use.
