中文摘要: 既往缺血性脑卒中的研究多从神经元的角度作为切入点,近年来对胶质细胞研究的不断深入,学者们逐渐意识到单纯研究神经元有一定的局限性。本项目是在前期研究工作的基础上,采用线栓法制备局灶性MCAO模型,采用Springbio高通量蛋白质芯片方法检测并筛选脑缺血后星形胶质细胞和神经元之间缝隙连接(GJ)相关特异性蛋白、Western-blot方法检测星形胶质细胞标志蛋白GFAP、神经元特有标志蛋白NF和轴突生长抑制蛋白Nogo-A、胶质细胞和神经元JAKs-SATA信号转导通路蛋白JAK1和SATA3不同时间段的表达变化,透射电镜观察突触数目及结构变化,探讨头穴丛刺法参与调控脑缺血后胶质细胞活化引起的相关蛋白质变化,从而发挥对神经元的保护作用,以期从分子水平揭示头穴丛刺法对脑缺血大鼠神经元保护的分子生物学机制,将为缺血性卒中的临床治疗提供新的思路。

英文摘要: In the past, the research on ischemic stroke was often studied from the point of view of the neurons. With the deepening research of glial cell, the limitation of the study on neuron gradually became awared of in recent years. On the basis of preliminary study ,The animal models are to be made by middle cerebral artery occlusion (MCAO) with the intraluminal filament. Springbio high-throughput protein chip method is to be used to detect the expression of astrocyte marker protein GFAP, the gap junctiongap junction related proteins,the neurons unique markers of protein NF and Nogo-A and of the JAKs-SATA signal transduction pathway proteins JAK1 and of SATA3 at different time segments.Transmission electron microscope is to be used to observe the ultrastructural changes of synapse number and structure.To explore the relative protein changes caused by glial cell activation regulated by cluster scalp acupuncture which play a protective effect on neurons after cerebral ischemia. It is expected to reveal the molecular biology mechanism of the protective effect on neurons after cerebral ischemia by cluster needing of scalp acupuncture. To provide new ideas and theoretical basis for the clinical treatment of ischemic stroke.
