中文摘要: 造成我国大规模养殖鱼类死亡的赤潮灾害主要是由鱼毒性赤潮生物引起,这类生物近年来已成为我国重要的有毒藻华原因种,其产生的溶血毒素是导致鱼类中毒死亡的重要原因。然而,目前对鱼毒性藻类及其溶血活性的识别仍十分困难,严重制约我国对赤潮灾害的有效管理。本项目以我国主要的鱼毒性赤潮藻类为研究对象,提取不同生长阶段鱼毒性藻类的溶血毒素并检测其溶血活性和叶绿素三维荧光光谱,用小波分析等方法提取鱼毒性藻类的荧光特征光谱, 运用费舍尔(Fisher)判别法建立识别鱼毒性藻类及其溶血活性的判别函数和判别函数图,并以不同生长环境及赤潮现场水体中的有毒和无毒藻类进行验证和完善;通过与无毒藻类色素的比较,结合鱼毒性藻类的荧光特征谱,研究产毒藻类的色素成分特征及其与溶血活性相对应的变化规律,探讨鱼毒性藻类荧光特征谱的物质基础,为弄清鱼毒性藻类溶血毒素的生物合成机理提供理论依据.

英文摘要: Ichthyotoxic algae blooms are ascribed to massive fish mortality in marine aquaculture in China. In recent years ichthyotoxic microalgae have turned to be the dominant causative species of HABs along the coast of China. These microalgae can produce hemolytic toxins, which are proved to be the main reason for fish killed. However, identification of hemolysin-producing microalgae and determination of toxicity of these organisms are quite difficult. In this project, we will study hemolytic activity and three dimensional fluorescent spectra of ichthyotoxic algae in different growth phase under laboratory conditions. Based on methods of wavelet analysis and Fisher discriminant analysis, we will extract the characteristic spectrum of three dimensional fluorescent spectra and investigate the correlation between characteristic spectrum of fluorescence and hemolytic activity. Furthermore, experimental verification of the correlation will be performed depending on variation of hemolytic activity and characteristic spectrum of fluorescence. In addition, we will study the characteristics of chromatophore under different environmental conditions and investigate the material basis for characteristic spectrum of fluorescence.
