中文摘要: 本项目是前一个结题的国家自然科学基金的延伸性研究。课题组在完成项目中,发现了值得进一步探索的新课题-温里中药的药性表征与TRPV1通路介导的UCP1调节热生成关系密切。基于此,本项目拟通过整体和离体相结合的研究模式,以伤害性热感受器TRPV1通道及其介导的产热关键节点-UCP1调节的热效应为新的视角,引入基因转染、基因敲除等关键适宜技术和方法,构建人TRPV1通道的异源表达体系和TRPV1基因缺失动物模型,结合TRPV1原位基因和野生型小鼠的生物效应的比对分析,以及基于寒证模型的以寒测热、以证测药(性)的反证研究,全面系统揭示温里中药表征温热药性的分子基础。同时,以UCP1热效应为基点,探索温里中药多成分、多靶点的药性整合机制和基于热偶检测技术的寒热药性评测体系。该项目旨在阐明中药药性表征的内在生物效应规律,进而丰富和完善药性理论内涵,为全面揭示中医药理论体系的现代生物学本质积累有效数据。

英文摘要: This project is a continual study of the finished project from National Natural Science Foundation. Our research team found a new area worthy of further exploration-the common molecular mechanism of the interior-warming herbs is closely related to TRPV1-mediated UCP1 heat-generating pathway. Based on this, the project will introduce gene transfection,gene knock-out technologies and methods to construct human-TRPV1-expressing system and TRPV1 gene-deletion animal model,together with the analysis of the biological effects demonstrated in in-situ-TRPV1 channel and wild-type mice,to comprehensively study the nature-demonstrating molecular basis of interior-warming herbs. Meanwhile, integration mechanisms of the multi-component and multi-target of interior-warming herbs around UCP1 thermal effects will be explored systematically. A thermocouple detection and analysis system that can evaluate the cold-heat natures of interior-warming herbs will also be established. The project aims to clarify the total nature-demonstrating characterization and common molecular mechanisms of the interior-warming herbs,thereby enrich and improve the theoretical meaning of Traditional Chinese Medicine (TCM) for the full exploration of the essence of modern biology of the TCM theory system.
