中文摘要: 浮游病毒是海洋中丰度最高的浮游生物,在海洋生态系统中有重要的功能,它们可以调控宿主的种群数量和群落结构,增加宿主基因组多样性。近年来两个国际性大规模海洋调查计划(GOS和Tara)都将浮游病毒作为重要的研究内容之一,认识到病毒基因在海洋生态系统中有广泛分布和丰富的多样性,并且还发现了许多新的病毒种类。目前我国海洋浮游病毒多样性研究不足,本项目拟在胶州湾采样,建立浮游病毒样品采集、制备的系统方法。利用透射电子显微镜技术和分子生物学技术,研究胶州湾浮游病毒的形态和遗传多样性,同时通过高通量测序技术开展浮游病毒宏基因组学研究,以了解胶州湾浮游病毒的优势种类及分布、种属组成等多样性信息。分析在不同季节、营养条件下胶州湾浮游病毒的多样性变化,比较胶州湾浮游病毒与其他海区在优势种类、种属组成上的异同。通过以上研究了解胶州湾浮游病毒的形态和遗传多样性,初步认识其在胶州湾生态系统中的作用。

英文摘要: Virioplankton are the most abundant plankton in the ocean. Marine viruses have been observed to modify, influence and control marine ecosystems. They can regulate host populations and community structure, and increase the diversity of the host genome. The two major international marine expeditation project (GOS and Tara) have investigated virus diversity in the ocean and found out that virus are extreme diverse. With several new kinds of viruses and hundreds of thousands of unknown gene sequences discovered in these projects, scientists now realize that virus are the biggest large and diverse pool of species in the world. The diversity of marine virus in China has been poorly studied. In this project, we focus on the diversity of virus in Jiaozhou Bay, China. Using transmission electron microscopy and molecular biology techniques to study the viral morphology and genetic diversity, the existence of dominant morphological and genetic-rich species, as well as the viral metagenomics would be expounded. Through these researches, an initial understanding of the role of virus in the Jiaozhou Bay ecosystem would be revealed.
