中文摘要: 芝麻枯萎病是一种分布广、防治较困难的世界性真菌病害,严重威胁和阻碍了芝麻的生产与发展。由于芝麻生产中缺乏突出的抗源材料,加之芝麻基础研究起步较晚、枯萎病抗性遗传规律尚不明确等原因,目前国内外芝麻抗枯萎病育种工作没有取得根本性突破。本研究拟首先利用Solexa测序技术开展不同抗/感芝麻材料在与枯萎病菌不同作用条件下的转录组表达谱研究,通过对抗感芝麻材料转录组表达差异进行系统的分析,以期全面了解芝麻在枯萎病病原菌侵染特定条件下的转录组基本表达模式,明确芝麻枯萎病抗性机理;其次,对于转录组获得的潜在抗枯萎病候选基因,本研究拟通过进一步对其在表达水平方面进行qRT-PCR验证和对其与188份芝麻自然群体在大田的抗枯萎病表型性状进行关联分析验证等研究,最终确定芝麻抗枯萎病目的基因和功能标记,为抢占芝麻基因资源、提升我国芝麻基础研究水平和利用分子手段培育高抗芝麻新品种奠定基础。

英文摘要: Sesame Fusarium Wilt (SFW) is an extinct and hard controlled fungus disease in the world, which restricts sesame production and development. Due to the lack of resistant materials and poor foundation for sesame basic research, the resistant genetic pattern of SFW was unknown, and no distinct breakthrough had been observed in sesame diseases resistance breeding. In the present study,in order to make a global survey of the transcriptome expression mode under the infection of Fusarium oxysporum and illuminate the fusarium wilt resistance mechanism in sesame, the transcriptome profiles will be analyzed between the resistant and susceptible sesame materials under different fusarium pathogen treatments with the Solexa RNA sequencing technology. Furthermore, qRT-PCR and fusarium wilt resistant association analysis with 188 sesame populations will be applied for evaluating the resistant candidate genes, and then disease- resistant genes and functional molecular markers would be validated, Based on the above research, we will gain more sesame gene resources for future sesame disease-resistance breeding, as well as accelerating China sesame fundamental research process.
