中文摘要: 大丽轮枝菌是一种土壤病原真菌,具有广泛的寄主。在我国,该病原菌侵染棉花所引起的黄萎病,因缺少有效的防治手段而难以控制。我们在前期的研究中发现,大丽轮枝菌的外泌蛋白VdCRP1是一个致病因子;而更有意思的是,在病原真菌和非病原真菌基因组中都存在VdCRP1的同源基因。为深入理解病原真菌致病性的进化演变规律,本项目将以VdCRP1为研究载体,以VdCRP1与寄主靶标的互作为突破口,探讨VdCRP1对大丽轮枝菌致病性的作用机理、以及与寄主靶标互作的特异性;并利用大丽轮枝菌VdCRP1敲除突变体菌株,比较不同真菌VdCRP1同源基因的功能,推测并验证VdCRP1蛋白的关键功能域和种间变异位点,揭示真菌外泌蛋白对病原真菌进化的可能作用模式。力求从致病因子与寄主靶标互作的特异性、以及外泌蛋白的变异规律阐述大丽轮枝菌的致病机理,为黄萎病的有效控制提供借鉴。

英文摘要: Verticillium dahliae is a soil-borne fungal pathogen that has broad host range. In China, it is a causal agent of cotton vascular wilt disease that causes large yield loss due to the lack of efficient controlling methods. In our previous study, a secreted protein VdCRP1 was found to be a virulence factor of V. dahliae. Interestingly, VdCRP1 homologous genes exist in not only pathogenic fungi but also non-pathogenic fungi. To further understand the evolution of fungal pathogens, we propose to investigate VdCRP1 for its mode of action on pathogenesis, and specificity of interaction between VdCRP1 and its host targets. In the meantime, VdCRP1 gene knock-out mutant would be used for functional comparison between VdCRP1 gene homologs to speculate and determine the key functional domains and mutational motifs, and therefore reflect evolutionary trends of fungal secreted proteins. This project aims to elucidate pathogenesis of V. dahliae with two aspects, specificity of effector - host target interaction and evolution of fungal secreted proteins. The results would be a reference for efficient control of vascular wilt disease.
