中文摘要: 酸敏感离子通道(ASICs)是一类由胞外酸化激活的阳离子通道,研究表明其在脑缺血、肿瘤等伴有局部组织酸化的病理过程中发挥重要作用。炎症常伴有组织酸化,ASICs是否参与了过敏性紫癜(HSP)的病理过程?我们前期研究发现HSP皮肤血管内皮细胞存在ASIC1a和ASIC3表达,药物抑制其表达对HSP血管损伤有保护作用。在此基础上,以人皮肤微血管内皮细胞(HDMVEC)为研究对象,分离HSP血清IgA1,制备HSP血管内皮细胞损伤模型,试图①运用全细胞膜片钳记录HDMVEC中H+激活或药物阻断的ASICs电流变化及电生理特点;②通过HDMVEC中ASICs过表达和表达沉默,观察ASICs对H+敏感性及HDMVEC生物学功能的影响;③用特异性和非特异性阻断剂阻断ASICs,观察其对HSP血管损伤的保护作用与机制。初步阐明HDMVEC中ASICs的生物学特性与功能,为HSP的防治提供新的治疗靶点。

英文摘要: Acid-sensing ion channels (ASICs) are a family of proton-gated cation channels activated by extracellular acid.Researches indicated that ASICs played an important role in the pathological conditions of brain ischemia,tumor and so on.Inflammation involved in tissue acidosis. Whether ASICs contributed to the pathological process of HSP has not been determined.Our previous studies showed ASIC1a and ASIC3 expression in skin vascular endothelial cells of HSP. In addition inhibiting their expression had a protective effect on injuried vessels of HSP.On this basis, we used human dermal microvascular endothelial cells(HDMVECs)as the research object,isolated serum IgA1 from HSP and made the model of injuried endothelial cells in HSP children to investigate: ①current changes and electrophysiological characteristics of ASICs activated by H+ in HDMVEC induced by IgA1 with cell patch clamp;②ASICs sensitivity to H+ and biological metabolism of HDMVEC by over expression and express silence of ASICs;③the protective effect on the injuried vessels of HSP by blocking ASICs with specific blockers and nonspecific blockers. We will firstly explain the physiological charateristics and function of ASCIs of HDMVEC and provide new strategies for preventing HSP.
