中文摘要: 茅苍术为燥湿健脾的常用中药,主产湖北大别山和江苏茅山地区,主要有效成分为苍术素、β-桉叶醇、苍术酮、茅术醇等萜类化合物。不同产地茅苍术成分的差异造成资源品质的不同,但目前尚缺乏系统生物学水平的资源品质研究。加上有效成分生物合成途径关键步骤萜类骨架的环化机制尚不清楚,极大限制了茅苍术资源开发利用及代谢调控的开展。课题组已建立了茅苍术种质资源圃、组织培养体系和挥发油GC/MS分析方法,本项目拟采用Solexa高通量转录组测序结合基于GC/MS技术的代谢组学方法,系统研究不同产地茅苍术的资源品质;通过构建基因表达谱和代谢谱,筛选负责萜类骨架环化的关键基因,再利用RACE及原核表达系统进行基因克隆及功能鉴定;最后利用qRT-PCR研究关键基因的表达模式。本研究结果将为研究植物资源品质提供新思路,为阐明茅苍术有效成分合成和调控的分子机制提供科学依据,并为优质新品种选育及资源创新、利用奠定理论基础。

英文摘要: Atractylodes lancea, mainly distributed in Dabie Mountain and Mao Mountain, is a commonly used medicinal plant, with terpenes as its principal effective component, such as atractylodin, atractylone, β-eudesmol, and hinesol. The quality of germplasm resources differs since the effective component varied in plants with different producing area. However, it is lack of study on the level of systems biology. Additionally, the molecular mechanism of effective component biosynthesis is still poorly understood, which severely impedes the study on germplasm resources usage and metabolic regulation. We have established germplasm resources garden and tissue culture system of A. lancea, and developed a GC/MS method for the component analyzing of essential oil. This project was designed to characterize the quality of germplasm resources of A. lancea from Dabie Mountain and Mao Mountain using Illumina Solexa high throughput RNA sequencing technology in association with metabolomic methods. The profiles of digital gene expression and metabolites will be established to screen the key gene for effective component biosynthesis. Finally, RACE, bacterial expression system, and qRT-PCR will be employed to characterize whole gene sequence, gene function and expression profile, respectively.
