中文摘要: 粘细菌细菌是极具研究开发价值的药源微生物新类群,其复杂的生活史和特殊的代谢机制已经引起科学家的重视。但受限于菌株的分离纯化困难,加上粘细菌在液体培养时不生长或者生长聚团,致使粘细菌的应用研究远远落后于其他的微生物类群。热带亚热带森林环境是分离粘细菌、发现新的粘细菌资源最好的来源。本项目以典型的热带亚热带地区原始森林和不同药用植物为研究对象,开展粘细菌的多样性和分离研究;探求筛选粘细菌助长菌和基于助长菌的粘细菌分离新方法,解决制约粘细菌分离的瓶颈,致力于获得未培养的粘细菌新资源,为粘细菌的分离提供新的思路;同时创新粘细菌液体培养技术体系,揭示不同的粘细菌菌株抗菌和抗肿瘤活性,分析活性产物的结构;本项目是首次开展热带亚热带原始森林和南药植物中粘细菌资源的分离和功能评价研究,极有可能得到遗传背景多样、功能新颖、有重要利用价值的粘细菌资源和活性产物,将为粘细菌应用研究提供更多的物质基础和技术支撑。

英文摘要: Myxobacteria is novel microbial phylum with high value of research and development for medical purpose. Its complex lifecycle and special metabolic mechanism attract the scientists' attention. However, due to the difficulties in isolation and purification of myxobacteria isolates as well as their inability to grow or aggregation in liquid culture, the applied study of myxobacteria lags far behind those of other microbial phylum.The tropical and subtropical forest environment is one the most appropriate resources of novel myxobacteria isolates. Targeting the typical tropical and subtropical primitive forests and medical plants, this study will isolate the myxobacteria and investigate the diversity. We will explore the helper bacteria (HB) of myxobacteria and establish novel technique for the isolation of myxobacteria based on HB, which can solve the bottleneck of myxobacteria isolation, aquire novel resource of unculturable myxobacteria and provide with new ideas for myxobacteria isolation. Meanwhile, we will create novel techniques for the liquid culture of myxobacteria, reveal the antibiotic and antitumor activity of different isolates and analyze the structure of active substance. This study is the first trial in the isolation of myxobacteria from tropical and subtropical primitive forests and medical plants, and in the eveluation of isolate functions, therefore, it is very likely that we will screen myxobacteria resources with diverse genetic background, novel function and important active metabolites. This can provide with more substantial basis and technique support for the applied sudy of myxobacteria.
