中文摘要: 了解和调查Streptococcus thermophilus代谢多样性及其功能基因对其发酵剂的研究、开发和进一步利用都具有非常重要的指导意义。本课题依托于内蒙古农业大学乳酸菌菌种资源库,以分离自传统发酵乳制品中的386株Streptococcus thermophilus为研究对象,参考前期多位点序列分型和产香特性关联分析结果,首次采用基因组重测序方法对具有不同产香特性菌株的代谢多样性及其功能基因进行全面系统的研究。在此基础上,利用PacBio SMRT测序技术完成代表性Streptococcus thermophilus菌株DNA甲基化图谱的破译,以期全景式的揭示其不同产香特性差异发生的分子机制。该项目的实施和完成可为Streptococcus thermophilus资源的利用及其发酵剂的开发奠定坚实的理论基础。

英文摘要: Understanding and investigation of gene diversity and metabolism of Streptococcus thermophilus are very important for starter development and further use. Based on 386 Streptococcus thermophilus strains isolated from the traditional fermented dairy products that have been preserved at the Lactic Acid Bacteria Collection Center at Inner Mongolia Agricultural University and the previous results from correlation analysis betwwen Multilocus Sequence Typing and studied aroma property, genome re-sequencing technology was applied for the first time to compare the metabolic pathways and functioanal genes responsible for aroma production in the strains. In addition, the DNA methylation maps of representative Streptococcus thermophilus strains will be decoded using PacBio SMRT sequencing technology to reveal the molecular mechanism behind the differences in their ability to produce aroma. The implementation and further completion of this project will lay a solid theoretical foundation for the utilization of Streptococcus thermophilus resources and the development of starter cultures.
