中文摘要: 生物质能源发展需要突破原料高效转化的瓶颈,以糖为转化目标进行能源生产是发展生物质能源的主要方式之一。挖掘优良基因、开展转基因培育新品种是生物质原料产能效率大幅度得以提高的重要路径。项目以割手蜜为研究对象,以甘蔗等近缘禾本科植物已克隆出的蔗糖磷酸合成酶基因为参考,设计引物,进行同源克隆,并检测基因在割手蜜体内的表达规律。获得的基因转化到烟草中验证基因功能,借鉴现有的甘蔗等近缘禾本科物种的组培和转基因体系,研制割手蜜的组培和转基因体系,开展基因转化及基因在割手蜜体内的调控特点研究。研究结果将是对目前蔗糖合成酶的研究的补充和丰富,获得的基因和技术将为多年生、高含糖量能源草的选育创制奠定基础。

英文摘要: Low efficiacy of biomass feed stock is the most limit factor for bioenergy. That sugar were traslated into ethonal is a relative easy way for bioenergy produce.It depends on finding the exlent gene and creating new vultivars by gene multifing. Saccharum spontaneum is a prenial and with high sugar content potential grass. In reference to other SPS genes which aready were cloned from other monocot,eg.sugarcane,we will design the fit primer to clone SPS gene from Saccharum spontaneum . In reference to other monocot trasformation systems,we will develop an eficient tranormation system for Saccharum spontaneum including tissue cultivate system.the expression characteristics ,the control ruler for sugar,starch and fiber of SPS gene of Saccharum spontaneum will be studied. the research result will be helpful for studies of SPS gene, the gene and technology havested from this project will be supplied to create new biograss cultivar aimed to high sugar content and prennial object.
