中文摘要: 先天性心脏病的发生机制十分复杂。表观遗传效应和基因遗传效应共同作用而导致先心病的发生。表观遗传效应最重要之一是DNA甲基化,它可影响多个基因的表达,与先心病密切相关。MicroRNA可影响多个基因的表达,并调节不同转录因子,从不同路径影响先心病发生。本研究从先心病病例出发,分析DNA甲基化水平变化和microRNA表达变化的相关性。在心肌细胞水平及斑马鱼整体水平,分别调节microRNA的表达和DNA甲基化水平高低,研究1)对心肌细胞或斑马鱼发育的影响和表型;2)microRNA的表达变化对DNA甲基化水平的影响及机制;3)DNA甲基化水平的变化对microRNA的表达的影响及机制;4)调节microRNA的表达是否可抑制DNA甲基化效应。由此,研究microRNA和DNA甲基化的相互作用在先心病中的作用,探索是否可通过调节microRNA的的表达抑制甲基化效应而达到干预先心病的发生。

英文摘要: Congenital heart disease (CHD) is a complex disease, with environmental and genetic factors playing important roles. The molecular mechanisms of CHD include genetics and epigetics. DNA methylation is one of most important epigenetic effect. Manay genes were involved in the signalling pathway of DNA methylation and microRNAs, with correlation with CHD. In this study, CHD clinlic sampls will be well collected. DNA methylation status, expression levels of microRNAs, and their correlation will be analyzed. In cultured myocardial cells or zebrafish in vivo, DNA methylation level or microRNAs expression level will be up- or down-regulated seperately,so as to study 1) effects of their change on differentiation and development of zebrafish cardiobascular system or cultured myocardial cells; 2) effect of microRNAs expression level on DNA methylation ststus; 3) effect of DNA methylation level on expression level of microRNAs; 4)wether manupilation of microRNA expression will inhibit DNA methylation effect. Overall, this study will expand our understanding of roles of microRNAs and DNA methylation on CHD, which will lead us to a better way to control heart development and to prevent CHD.
