中文摘要: 肿瘤微环境对肿瘤的演进和转归起重要作用,肿瘤浸润淋巴细胞(TILs)是肿瘤微环境中的重要组成成分。我们的前期研究发现不同分子分型的乳腺癌微环境中TILs的数量、类型、比例及分布等均存在明显差异。本课题将进一步通过大样本临床数据分析以及体外模拟肿瘤微环境,对比研究不同分子亚型乳腺癌中的TILs,拟采用组织芯片、流式细胞技术、免疫磁珠细胞分离、体外三维立体细胞培养、实时动态细胞摄像、ELISA等方法,分析不同分子分型乳腺癌中TILs的功能状态和分子机制,分析其与临床病理学特征、预后及治疗的关系,寻找针对不同分子分型乳腺癌的预后预测和免疫治疗靶点,特别是为无法从激素受体或HER2/neu靶向治疗等乳腺癌重要的后续治疗中获益的分子亚型提供免疫靶标。

英文摘要: The tumor microenvironment plays an important role in the progression of breast cancer, and tumor infiltrating lymphocytes (TILs) is one of its important components.In our preliminary studies, we have identified that TILs in different molecular subtypes of breast cancer have remarkable differences in their number, type, proportion and distribution.This project is going to expand the prior study to clinical analysis of large tissue samples and assessment of TILs in in-vitro tumor microenvironment. The comparative study is going to employ tissue microarray, flow cytometry, immunomagnetic bead cell separation, in vitro three-dimensional cell culture, real-time dynamic camera cell and ELISA, to assess the functional status and molecular mechanisms of TILs in different subtypes of breast cancer, to identify their relationship with clinico-pathological features, prognosis and treatment options, to look for predictors for prognosis and for immune therapeutic targets, especially for those subtypes that can not benefit from endocrine therapy and HER2/neu target therpay due to lack of hormone receptors and/or HER2/neu expression.
