中文摘要: 维生素C(VC)作为一种重要的抗氧化剂,在水产养殖中一直被广泛应用。然而近年来越来越多的研究表明,在一些水产动物中补充大量VC并未增强机体的抗氧化能力,甚至还会降低机体的抗氧化能力,表现出促氧化作用。目前,VC对水产动物抗氧化能力影响的研究很多,但多停留在细胞和亚细胞水平的现象描述,VC对水产动物氧化损伤影响(抗氧化/促氧化)的分子机制未见报道。因此,本项目拟以我国重要淡水养殖经济鱼类泥鳅为研究对象,首先探明投喂不同剂量VC对饲料氧化鱼油胁迫下泥鳅肝脏氧化损伤的影响。接着采用RNA高通量测序技术和生物信息学分析方法比较分析氧化损伤泥鳅VC缺乏组、VC适量组和VC过量组间鱼体肝脏的基因表达谱,然后筛选、克隆关键基因并进行基因表达和调控分析,最终阐明VC对泥鳅肝脏氧化损伤影响的分子机制。本项目将填补VC对水产动物作用的分子机制研究的空缺,同时为VC在水产养殖上安全有效地利用提供重要的科学依据。

英文摘要: The biological activity of vitamin C (VC) is frequently considered to be an important anti-oxidant, which is wildly used in aquaculture. However, more and more studies have recently been demonstrated that a large dose of VC supplementation did not improve antioxidant capacity of aquatic animals. Moreover, it may decrease the antioxidant capacity of aquatic animals as pro-oxidant. At present, many studies have been indicated the anti-oxidant effects of VC in aquaculture, but most of those studies just described the results at the cellular and subcellular level. The molecular mechanisms of anti-oxidant or/and pro-oxidant capacity of VC remain unclear in aquatic animals. Therefore, this project will choose Misgurnus anguillcaudatus, an important freshwater commercial fish, as the experimental object. Effects of different dose of VC supplementation on growth performance, oxidative condition of liver and body of juvenile loach feeding with oxidized fish oil will be firstly estimated. And then, we will compare the gene expression profiles among livers of VC deficiency, VC optimum and VC excess groups based on the RNA-seq high-throughput sequencing technology.
