中文摘要: 线粒体是细胞里面的能量工厂,也参与了细胞凋亡的过程。线粒体及时的更新对于维持线粒体的整体功能十分重要,其主要依赖于线粒体生物发生和线粒体自噬,两者分别控制着线粒体的数量和质量,而对于两者之间关系的研究很少。FUNDC1是我们鉴定的新的线粒体自噬受体,我们发现FUNDC1的启动子区域有线粒体生物发生相关的转录因子NRF1的结合位点。NRF1敲减后FUNDC1的蛋白表达量明显下调。NRF1的转录辅激活因子PGC1α也能上调FUNDC1的表达水平。寒冷刺激后小鼠棕色脂肪组织中的PGC1α的表达上调也伴随着FUNDC1表达的上升。我们将进一步研究NRF1对于FUNDC1的转录调控,同时明确PGC1α对于FUNDC1的调控是否依赖于NRF1,并在动物模型上研究FUNDC1在线粒体生物发生及线粒体自噬中的功能。我们的研究将有助于揭示线粒体质量和数量控制之间的相互调控关系及生理意义。

英文摘要: As the cell's powerhouse, mitochondria also play a central role in the control of apoptosis.Mitochondrial function is tightly regulated by mitcondrial turnover. Although mitophagy and mitochondrial biogenesis play important role in the regulation of mitochondrial quality and quantity,there are few studies about the relation of these two processes. FUNDC1, which is a new mitophagy receptor,had been identified by our lab recently. Our study revealed that there is a putative NRF1 binding site in the promoter of FUNDC1. The protein level of FUNDC1 is also down regulated in NRF1 knockdown cells.As a coactivator of NRF1,PGC1α also can elevate the expression of FUNDC1 both in vivo and in vitro.We will investigate the transcriptional regulation of FUNDC1 by NRF1 and PGC1α and elucidate the biological relevance of FUNDC1 in mitophagy and mitochondrial biogenesis. Our study will provide new insight about mitochondrial quality and quantity control.
