中文摘要: 宫颈癌的发病率居妇女恶性肿瘤第二位,人乳头瘤病毒(HPV)感染与宫颈癌的发生密切相关。我们用蛋白质组技术发现RbAp48是HPV感染的宫颈细胞癌变的关键调控蛋白,研究揭示RbAp48是放射敏感性蛋白。本项目通过研究宫颈癌细胞Caski、动物肿瘤模型及患者病变组织放射前后RbAp48的表达差异来确定其是否是放射敏感性蛋白;在宫颈永生化细胞H8抑制RbAp48后行放射处理,分析抑制RbAp48是否降低细胞的放射敏感性,用Caski增强RbAp48表达后再行放射处理,分析过表达RbAp48是否提高细胞的放射敏感性;构建稳定表达RbAp48的重组腺病毒载体,感染Caski致瘤的裸鼠,结合放射处理,观察肿瘤生长,从体内试验揭示RbAp48能否提高放射敏感性。本项目首次将RbAp48结合放射用于治疗宫颈癌的研究,可望为临床上放疗加基因协同治疗宫颈癌奠定基础,为宫颈癌治疗开辟新径,有重要的实际应用前景。

英文摘要: Ionizing radiation has been used to treat cancers for a century. However, radioresistance remains a major problem in the clinic. Etinoblastoma-binding protein 4 (RbAp48) was also isolated as one of three radiosensitive genes in a microarray analysis used for selecting radiosensitivity prediction molecules and RbAp48 overexpres- sion-induced radiosensitization in HS-578T, MDA-MB-231, and MALME-3Mcells, two breast cancer and amelanoma cell line, respectively, when compared with mock-transfected cell lines. Our Previous study have showed that RbAp48 was a novel critical mediator in cervical cancer. We hypothesized that cervical cancer cell lines exhibiting radioresistance show signi?cant upregulation of RbAp48 and inhibiting RbAp48 would suppress tumor cell radioresistance and provide a new treatment approach for human cervical cancer.
