中文摘要: 尽管人们对小RNA在人和动物病原细菌致病过程中的调控作用已有一些了解,但对小RNA在植物病原细菌致病过程中的作用还知之甚少。到目前为止,在十字花科黑腐病菌(Xcc)-一个研究微生物与植物相互作用的模式菌中还没有小RNA参与致病的报道。为了阐明小RNA在Xcc致病中的作用,近年来,我们在Xcc中开展了小RNA鉴定和功能研究工作。我们发现,过量表达小RNA sRNAXCC-24导至Xcc致病力显著下降但不影响其在基本和丰富培养基中生长,说明该小RNA在致病中起重要的调控作用。在本申请项目我们将(1)鉴定Xcc中所有受sRNAXcc-24调控的基因并确定其中的致病基因,(2)鉴定sRNAXcc-24的直接调控靶标以及它在靶标mRNA上的结合位点和结合靶序列,(3)弄清sRNAXcc-24的结合对靶标基因表达的影响,以达到阐明sRNAXCC24调控致病的分子机理的目的。

英文摘要: Although the regulatory role of small RNA (sRNA) in the pathogenesis of human and animal bacterial pathogens has been studied in some detail, little is known about the role of sRNA in the pathogenesis of plant bacterial pathogens. Thus far, no sRNA has been assigned a definitive role in the virulence of the crucifer black rot pathogen Xanthomonas campestris pathovar campestris (Xcc), a model bacterium for studying microbe-plant interactions. To understand the role of sRNA in Xcc virulence, in recent years we have been carrying out identification and functional analysis of sRNA in Xcc. We found that over-expression of the small RNA sRNAXcc-24 resulted in reduced virulence of Xcc but no effect on its growth in rich and minimal media, indicating that the sRNA plays an important regulatory role in the pathogenesis of Xcc. To clarify the molecular mechanisms by which sRNAXcc-24 regulates the pathogenesis of Xcc, in this project, we plan to (1) identify all genes and virulence genes that are regulated by sRNAXcc-24; (2) identify the direct targets of sRNAXcc-24, and the binding site as well as binding sequence of sRNAXcc-24; (3) determine the affects of sRNAXcc-24 binding on the expression of its target genes.
