中文摘要: 人表皮生长因子受体过表达存在于乳腺癌等许多恶性肿瘤的细胞中,明确该受体表达状态对于乳腺癌的诊断、靶向治疗和预后评价至关重要。尽管基于Gd-DTPA的动态增强MRI技术已成为乳腺癌的重要检查手段,但仍存在特异性和敏感性不高等问题。为了解决该难题,基于配体-受体反应特异性的原理,以课题组前期关于肿瘤分子靶向性药物和受体靶向型造影剂前期研究为基础,将钆配合物Gd-DO3A与配体活性肽(CMYIEALDKYAC,EBP)偶联,合成出新型受体靶向性对比剂EBP-Gd-DO3A。本课题拟开展体外细胞EBP-Gd-DO3A结合试验、结合抑制试验、磁共振成像试验和磁共振成像抑制试验,以及动物体内急性毒性试验和肿瘤磁共振成像试验。旨在进一步探讨该对比剂的作用机理、寻靶能力及组织成像能力,为评价该对比剂用于活体肿瘤成像的可行性提供依据,也为构建能提高肿瘤成像特异性和敏感性的对比剂打下基础。

英文摘要: Human epidermal growth factor receptor is overexpressed in breast cancer and other epithelial malignancies, which is very importment in diagnosis, targeted therapies and post-clinical evaluation of breast cancer. Although magnetic resonance imaging (MRI) based on gadolinium complex has become the tool of inspection of breast cancer, it is still non-specific and low-sensitive. To solve the problems, this study plans to use ligand active peptide (CMYIEALDKYAC;EBP) conjugated with Gd-DO3A and compose a new kind of contrast agent EBP-Gd-DO3A. Here the topic is to discribe the mechanism, targeting ability and imaging capabilities of the novel tagagent from molecular levels, cell levels and animal experiments. The aim of the project is to construct the contrast agent which can improve the specificity and sensitivity for tumor imagine of MR, also to provide the research data for the feasibility of in vivo tumor imaging.
