中文摘要: 溶剂显著地影响分子的荧光发射。为了获得分子的真实信息,人们开始尝试采用电喷雾电离生成的气态离子研究分子荧光,但该领域的研究刚刚开始,仍有许多问题需要解答。为此,本项目拟研制电喷雾电离-荧光联用装置,并利用常规荧光染料如罗丹明等对该装置进行性能表征。研究对象选择荧光标记的适配子,通过比较研究其气、液两相的荧光性质,从理论和实验两个方面阐述气相中适配子的荧光特性,进一步研究荧光染料之间及G碱基与染料之间气相中的荧光共振能量转移,探讨荧光发射光谱、吸收光谱和疏水作用力的改变对气相中的荧光共振能量转移效率的影响。在此基础上,阐明荧光和吸收性质改变及溶剂效应在荧光共振能量转移中的作用。本项研究将研制气相中荧光分析的新装置,建立研究气相中荧光共振能量转移的新方法,探讨适配子气相中的荧光性质和荧光共振能量转移,获得物质分子荧光发射的本质特征及有关气相中荧光共振能量转移的理论问题。

英文摘要: In liquid phase, solvent dramatically influences the molecular fluorescence. With the purpose of obtaining the true information, reseachers started to study the molecular fluorescence with gasous ions produced by electrospray ionizatio(ESI).However, this research is just at the beginning and many questions remain to be explored. In this proposal, a new ESI-fluorescence device will be constructed and the performance of this new device will be characterized by common dyes such as rhodamine. Choosing fluorphore-labeled aptamer as the reseach object,the fluorescence of aptamer in liquid and gas phase will be compared and then the fluorescence properties in the gas phase will be explored by therory and experiments.Further, the fluorescence reasonance energy transfer (FRET) of dye-dye and guanine-dye in the gas phase will be investigated to explore the influences to the emission spectra, absorption spectra, and hydrophobic interaction changes on FRET efficiency.On the basis of these,the role of fluorescence and absorption property changes and solvent effect on FRET in the gas phase will be carefully elucidated.
