中文摘要: 包括糖尿病在内的代谢综合征的发病率不断升高,其中胰岛素抵抗(IR)起核心作用。Vaspin是最近发现的一种脂肪细胞因子,具有激素和细胞因子作用,可以降低IR,但其机制尚不清楚。Vaspin既可以直接影响胰岛素信号通路,也可以间接影响该通路的信号转导分子(JNK、IKKβ和NF-kappaB等)。在我们前期工作基础上,本项目利用western blot,real time PCR、基因转染、流式细胞检测、免疫荧光染色以及RNAi等技术,检测Vaspin在肝细胞、人脐静脉内皮细胞和脂肪细胞中诱导的信号分子和细胞功能(糖原生成,葡萄糖转运,粘附分子表达等);并通过干预手段(激动剂、抑制剂或siRNA等)与动物模型相结合的策略,在个体和细胞水平上确定Vaspin与胰岛素抵抗之间的相互关系。本项目的完成将为与Vaspin有关的代谢综合征的治疗和预防提供有实质价值的理论依据。

英文摘要: The incidence rate of the metabolic syndrome, including diabetes, had continuously increased and insulin resistance (IR) plays a central role in it. Vaspin is a recently discovered adipokine with both hormone and cytokine functions to reduce the IR; however, its mechanism is unclear. Vaspin could affect either through a direct insulin signaling pathway or indirectly via the signal transduction molecules, such as JNK, IKKβ, and NF-kappa B, etc. Based on our preliminary work of this project, we used western blot, real time PCR, gene transfection, flow cytometry, immunofluorescence staining and RNAi technology to detect Vaspin-induced signaling molecules and cell function (glycogenesis, glucose transporter, expression of adhesion molecules, etc.) in liver cells, human umbilical vein endothelial cells and fat cells; using interventions methods (agonists, inhibitors, or siRNA, etc.) and together with animal models combining strategies to determine the relationships between the Vaspin and insulin resistance at the individual and cellular level. Accomplishments of this project will provide a practical, valuable and theoretical basis for the Vaspin-related metabolic syndrome treatment and prevention.
