中文摘要: 由于适应性免疫的关键细胞和分子只在脊椎动物中被发现,因此人们一直认为无脊椎动物不具有典型的适应性免疫应答,天然免疫承担其抵抗外来病原侵袭的主要功能。然而,我们前期的研究在脊索动物文昌鱼中发现了淋巴样细胞及淋巴细胞信号转导的关键功能分子,并找到了与适应性免疫相伴而生的外源性细胞凋亡信号通路。最近我们又从文昌鱼基因组发现了RAG1/RAG2的同源基因,这些证据提示我们原始的适应性免疫应答可能已在脊索动物中存在。因此,本项目拟在前期研究的基础上,进一步在文昌鱼中追踪鉴定免疫相关的细胞及组织,研究文昌鱼淋巴样细胞中是否存在RAG1/RAG2及DNA聚合酶X家族成员所介导的重排修复机制,从而揭示抗原受体多样性产生机制的起源,为重新理解适应性免疫系统的起源和演化提供直接的证据。

英文摘要: Since the hallmarks of vertebrate adaptive imunity could not be found in invertebrates,innate immunity was generally thought to be the major defense system of invertebrates. However, in our previous studies, lymphocyte-like cells and genes involved in lymphoid signaling have been identified in the basal chordate amphioxus. Moreover, the homologs of RAG1 and RAG2 were recently been cloned from amphiouxs. These evidences suggest that the primitive adaptive immunity may has exsited in amphioxus. Thus, further identification of the immune-related cells and seeking the RAG1/RAG2 mediated recombination in amphioxus lymphocyte like cells, should provide direct evidences for understanding the origin and evolution of vertebrate adaptive immunity.
