中文摘要: 血管生长正负调控因子的平衡及转换调控血管新生,其对肿瘤的生长和转移至关重要。我们发现,血管生长负调控因子TNFSF15可显著下调血管内皮细胞VEGF的分泌,抑制血管新生和肿瘤生长;而血管生长正调控因子VEGF能抑制血管内皮细胞TNFSF15的表达,促进血管新生、肿瘤生长和转移。MicroRNA在TNFSF15和VEGF相互调控中发挥重要作用。本项目旨在揭示TNFSF15与VEGF如何通过microRNA相互钳制以及对肿瘤血管新生调控的机制,有望成为治疗肿瘤的候选者。

英文摘要: Tumor growth and metastasis rely on angiogenesis, which depends on switching the balance of positive and negative regulating factors. We recently discover that TNFSF15, a negative regulating factor for angiogenesis, can down-regulate the secreting VEGF of vascular endothelial cells, which inhibits angiogenesis and growth of tumor. However, VEGF, a positive regulating factor for angiogenesis, inhibits the expression of TNFSF15 in vascular endothelial cells, which promote angiogenesis, growth, and metastasis of tumor. MicroRNAs play an important role in antagonistic mechanism of TNFSF15 and VEGF. We will focus on researching that TNFSF15 and VEGF how to regulate each other by microRNA, and switch the tumor angiogenesis, which maybe discovery drug candidate for treat cancer patient.
