中文摘要: 《晶珠本草》称"沙棘活血化瘀,利心脏血脉"。沙棘黄酮制剂临床上用于治疗缺血性心脏病。氧化应激可导致重要生物大分子的损伤性氧化修饰,通过不同的信号转导通路引起细胞功能和结构的改变,最终导致细胞死亡。因此,寻找安全高效的抗氧化剂对抑制心肌细胞损伤的发生以及缺血性心脏病的治疗具有十分重要的意义。申请人在前期研究中发现,沙棘的抗氧化力主要来自抗坏血酸,其次为原花青素,黄酮的贡献率很小。本项目拟采用多元指纹图谱与多指标药效评价相结合的研究模式,运用数理统计手段进行谱效关系分析,探讨沙棘化学成分与抗氧化性及心肌细胞损伤保护作用三者之间的相互关系,揭示传统藏药沙棘对心肌细胞损伤保护作用的物质基础。通过配伍组分与多重药效指标的相关性分析,定量研究沙棘的黄酮/原花青素/有机酸组分之间的协同增效作用,寻找药效最优的配伍组合,为研究不同类型抗氧化剂的联合应用,发现新的天然组合药物提供基础性研究数据。

英文摘要: Hippophae fructus Hippophae fructus known as sea buckthorn berries is a traditional Tibetan medicine which was stated in shel gong shel phreng for "activating blood circulation to dissipate blood stasis thus benefiting heart and vessels". In clinical practice, the preparations of sea buckthorn flavonoids have been used for treating ischemic heart disease. It has been proved that oxidative stress can lead to injuring oxidative modification of important biological macro-molecules, which causes the changes on cell functions and structure through various signal transduction pathways, and finally results in cell apoptosis. Therefore, it is of significant value to seek safe and efficient antioxidants for inhibiting cardiomyocytes from injury and curing ischemic heart disease. From previous research, the applicant found that the antioxidant ability of Hippophae fructus attributes mainly to ascorbic acid, followed by proanthocyanidins, whereas flavonoids have little contribution. In this proposal, a research model combining multiple chromatographic fingerprints with multiple index activity evaluations will be employed to investigate the correlations between chemical compositions of Hippophae fructus against the antioxidant ability and the protection of cardiomyocytes injury, and reveal the material basis of this protect
