中文摘要: 唐氏综合症黏附分子(Dscam)及相关蛋白是一大类具有多个结构域 的细胞表面受体分子,广泛参与多个生物学路径,尤其在神经发育、生长及网络形成等方面起着重要作用,同时也与包括唐氏综合症在内的多种疾病有着直接的关系。Dscam及相关蛋白的膜外结构域通常都在十个以上,提示了其结构和功能的复杂性,同时也给结构研究带来了较大的困难。目前的研究结果表明,这些结构域可以形成复杂的构象,在配体识别和信号传递方面起着关键作用。但由于技术手段等方面的限制,与此相关的分子机制还知之甚少。本申请的目标,就是在已有的工作基础上,运用多种结构和分子生物学方法,尤其是通过结合蛋白质结晶学和电镜三维重构技术,获得Dscam及相关蛋白和复合物的高分辨三维结构,以及它们在脂膜上和体内的原位(或接近原位)结构信息,阐释其在神经发育、细胞生长与迁移过程中的分子机制。

英文摘要: Down syndrome adhesion molecule ( Dscam) and related proteins are multi-domain cell surface molecules involved in many biological pathways, especially neuronal development and growth, and also associated with many diseases.The multi-domain structure of Dscam and related proteins suggests the complexity of their structures and functions, which also make the structural studies difficult. Current data show that these proteins can have intriguing conformations to regulate the recogniztion of ligands and signal transduction. However,due to the technical limitation, the molecular mechanism of Dscam and related molecules are largely unknown. Here, we plan to investigated the high resolution structures of these proteins, and also the conformation of these molecules on the surface of membrane, both in vitro and in vivo,by combining X-ray crystallography and 3D reconstruction of electron microscopy, thereby elucidating the mechanisms of these molecules in neuronal development,cell growth and migration.
