中文摘要: 小反刍兽疫是由副粘病毒科麻疹病毒属小反刍兽疫病毒引起的一种急性、热性、高度接触性传染病,被OIE列为法定报告的A 类传染病,我国将其列为一类动物疫病,发病率和致死率极高,危害巨大。2007年我国首次报道在西藏发生了PPR疫情,严重的威胁了我国的动物卫生安全。目前关于该病毒入侵宿主过程中融合机制方面的研究还是空白。本项目拟通过分子模拟解析附着蛋白(血凝素蛋白)与受体复合物的结构,并以受体SLAM为主研究其与病毒附着蛋白、融合蛋白在融合过程中相互作用的分子机制,确定病毒与受体相互作用的关键位点,阐明小反刍兽疫病毒的融合机制。这将明确小反刍兽疫病毒致病机制,并为PPRV新型重组疫苗和抗病毒药物的研制奠定坚实的基础。

英文摘要: Peste des petits ruminants (PPR) is an acute, highly contagious and economically important viral disease in small ruminants, especially goats and sheep. PPR virus (PPRV) is a member of the genus Morbillivirus, family Paramyxoviridae. PPR is one of A class infectious diseases by OIE, and one of I type animal epidemic diseases in china. PPRV infection was officially reported in the Ngari region of western Tibet of our country in July 2007. It is a great threat to animal health in our country. At present reports of the fusion mechanism in the invasion process is rare.This project aims to resolve the structures of receptors in a complex with PPRV attachment (hemagglutinin) protein (PPRV-H) by molecular modeling, and focuses on the molecular mechanism of SLAM interaction with PPRV-H and PPRV fusion protein (PPRV-F) in the fusion process. We will determine the key sites of virus interaction with its receptors, and clarity the fusion mechanism of PPRV. The results will provide the evidence for understanding of viral pathogenic mechanisms, development of novel recombinant vaccines and antiviral drugs.
