中文摘要: 本课题针对喉部损伤,研究组织工程支架的外在几何形貌和内在构造与成分对喉软骨细胞粘附与生长的影响规律。基于患者MRI和CT医学影像,重构支撑喉的三维内外轮廓和生物物理结构的客户化、个性化带运动关节的3D软骨数字模型。采用兼具保护材料生物活性和复杂空间形状成形能力的低温沉积成形(LTDF)和弹性材料直接成形(EPMDF)方法搭建多元素优化喉软骨组织重建特定微环境科学平台,探索支架内在孔洞和形貌、种子细胞与营养因子等多元素对喉软骨细胞增殖和分化的影响机制。解决一般组织工程支架难以还原喉复杂空间形状、难以设计内在孔隙结构的问题。将DCW、EPMDF成形的喉软骨支架进行对比研究,构成具有几何分级结构、材料梯度的一体化喉损伤修复支架。探索高仿生支架对于喉软骨组织的生长、上皮细胞的爬行与粘附、支架降解与细胞再生的规律,并为探索组织工程化人工喉的走向临床奠定理论和实验基础。

英文摘要: This proposal focuses on the study on the influences of the outer geometry appearance and inner structure and component of the tissue engineering scaffold of laryngeal damnification on the conglutination and growth of larynx cartilage cells.Based on the MRI and CT images of the patient, the 3D digital model with the customized biological and physical structure will be reconstructed. A scientific platform for the optimization of the reconstruction of the larynx cartilage tissue with the low temperature diposition forming(LTDF) and elastic partical material direct forming(EPMDF) with the protection of biomaterial will be built to explore the influences of the porous shape of the scaffold, seed cell and nutrition factors to the multiplication and differentiation of larynx cartilage cells, and solve the problems that current tissue engineering scaffolds are difficult in the revivification of the complex shape and in the design of the inner porosity. Through comparing the scoffolds by LTDF and EPMDF methods, the integrative scaffold for the repair of larynx damnification with geometry hiberarchy and material grads will be constructed.
