中文摘要: 鸭瘟是由鸭瘟病毒(DPV)引起的鸭、鹅和其他雁形目禽类发生的一种急性败血性传染病,死亡率可高达80%。鸭瘟强毒感染可造成中枢免疫器官和组织的损伤及免疫抑制;病毒侵入机体后可产生潜伏感染,病毒长期存在于三叉神经节内,持续向外排毒。由于鸭瘟病毒感染之后引起的免疫抑制和潜伏性感染,使得鸭瘟的防治变得愈加复杂。本研究拟以DPV囊膜糖蛋白gD为目标蛋白,采用免疫共沉淀、GST Pull-down技术,筛选gD的细胞结合受体;通过受体阻断和受体重建实验确证所筛选的细胞结合受体;以生物信息学预测及同源病毒研究结果为基础,研究gD与受体相互作用的结合位点,构建结合位点突变体并转染宿主细胞,辅以流式细胞术及细胞融合实验,验证gD与gD受体结合位点,从分子水平上初步阐明鸭瘟病毒与宿主细胞作用的机制,该项目对深入研究鸭瘟病毒的致病机制,研制高效的病毒疫苗、抗病毒药物和诊断试剂,制定有效的防治措施具有重要意义。

英文摘要: Duck plague is an acute, contagious disease of ducks, geese and other Anseriformes animals caused by duck plague virus (DPV), the mortality rate can be as high as 80%. Infection of virulent Duck plague Virus can cause damage of central immune organ and tissue and immune suppression; invading of duck plague virus can produce latent infection, the virus persist in the trigeminal ganglion, discharged active virus into the environment for a long time. Because of immunosuppression and latent infection induced by duck plague virus, the prevention and control of duck plague becomes increasingly complex. This study focus on DPV envelope glycoprotein gD as a target protein for screening gD cell binding receptor by using co-immunoprecipitation, GST Pull-down technology. The selected cell binding receptor was confirmed by receptor blocking and screening receptor reconstruction experiments . Based on the bioinformatics predictions and findings of homologous virus , predicted gD receptor interaction binding sites ,constructed binding sites mutant and transfected into the host cell, supplemented by flow cytometry and cell fusion experiments to verify gD and gD receptor binding sites.
