中文摘要: H9N2亚型禽流感病毒(AIV)感染产蛋鸡群可引起产蛋率严重下降,产出畸形蛋、砂皮蛋和软壳蛋,持续时间长达1~2个月,引发严重的经济损失。但H9N2 AIV感染引起产蛋鸡产蛋异常的机理并不十分清楚。我们发现H9N2亚型AIV感染鸡输卵管上皮细胞(OECs)可引起细胞病变及凋亡。本研究拟将H9N2 AIV的NS1基因转染鸡OECs,研究H9N2 AIV的NS1蛋白在鸡输卵管上皮细胞中的亚细胞定位和NS1引起细胞病变和凋亡作用的主要功能区,探明鸡输卵管上皮细胞中与AIV NS1直接相互作用的胞内蛋白,明确鸡OECs内与禽流感病毒感染起关键作用的蛋白质分子。揭示NS1蛋白致鸡输卵管上皮细胞凋亡的途径。探索H9N2 AIV诱导鸡输卵管上皮细胞病变的分子机制,揭示H9N2禽流感病毒引起产蛋鸡产蛋异常的机理。研究获得的成果还可以为H5N1禽流感防控提供新的思路,具有重要的科学意义和潜在的应用前景。

英文摘要: H9N2 avian influenza virus (AIV) infection in the layers can cause serious decline in egg production, produce deformity eggs, sand preserved eggs and soft-shell egg,and this situation can last 1-2 months, causing serious economic loss. However, the pathogenic mechanism of H9N2 AIV infection is not clear. We found that H9N2 AIV infection could cause cells lesions and apoptosis in chicken oviduct epithelium cells(OECs). The determination of this experiment is to research the sub-cellular localization of NS1 protein of H9N2 AIV in Chicken OECs and the main functional region that NS1 causes cell lesions and apoptosis by transfecting NS1 gene into chicken OECs, to explore intracellular protein that directly interact with AIV NS1 in chicken OECs, to verify proteins that take key role in AIV infection in chicken OECs and to reveal the approach that NS1 protein causes cell apoptosis of Chicken OECs.Through this study,we expect that we can explore the molecular mechanism of pathological lesions in chicken OECs induced by H9N2 AIV and reveal the mechanism of egg abnormally in layers caused by H9N2 AIV. The results can also provide new ideas for prevention and control of H5NI avian influenza which has important scientific significance and potential application prospect.
