中文摘要: 生育三烯酚是维生素E的一种,具有调节免疫应答,抑制多种肿瘤细胞增殖且不影响正常细胞生长的作用。本课题组前期实验证实,富含生育三烯酚的棕榈油能抑制裸鼠结肠癌生长,δ-生育三烯酚能诱导人结肠癌细胞SW620程序化死亡。本项目拟通过化学诱导剂建立原位结肠癌动物模型,从大体观察、细胞形态学检测和DNA变化三方面研究δ-生育三烯酚的肿瘤增殖抑制作用;采用经典免疫方法探讨δ-生育三烯酚对动物的免疫增强作用;结合Western blotting、RT-PCR等分子生物学实验技术,研究T淋巴细胞、NK细胞、巨噬细胞、体液免疫应答的肿瘤杀伤作用、NF-κB信号通路诱导肿瘤细胞凋亡作用和肿瘤细胞的免疫逃逸现象,探讨δ-生育三烯酚肿瘤增殖抑制作用机制。本研究为开展δ-生育三烯酚对结肠癌的早期干预,增强人群免疫水平提供有力的科学依据和理论支持。

英文摘要: Tocotrienol is one of subtypes of Vitamin E,which can regulation of the immune response and specifically inhibit proliferation of cancer cells without bad affecting at normal cell growth. Our research group has demonstrated that tocotrienol-rich fractions from palm oil and δ-tocotrienol could significantly inhibit the growth of colon cancer in nude mice and the proliferation of human colon cancer cells SW620. To study the effect of δ-tocotrienol on proliferation of colon cancer by general observation, cell morphology observed and DNA changes; investigate the immune enhancement result with the classic immunological methods; combined with Western blotting , RT-PCR and other molecular biology techniques to study the effect of T cells, NK cells, macrophages and humoral immune responses on killing tumor, tumor cell apoptosis induced by NF-κB signaling pathway and tumor cell immune escape phenomenon. Investigation tumor growth inhibition mechanism of δ-tocotrienol . These data may provide a theoretical support and scientific basis of human colon cancer early intervention, and a strong boost population immunity levels.
