中文摘要: 骨髓间充质干细胞是一种多向分化潜能的成体干细胞,是目前最具应用优势的种子细胞。Myocardin相关转录因子家族是最新发现的血清反应因子(SRF)的协同转录因子,它们与SRF协同作用调控平滑肌细胞特异性marker的表达,是平滑肌细胞的发育和分化中所必需的辅助因子。本研究在先前研究Myocardin和TGF-β35843;节骨髓间充质干细胞向平滑肌细胞分化的基础上,进一步利用细胞和分子生物学的研究手段,证明:TGF-β26159;通过激活myocardin家族诱导骨髓间充质干细胞分化;TGF-β20449;号通路下游smad与mycardin之间的相互作用及作用位点;二者的协同作用及作用机制;周期蛋白p16和Rb的作用;p38、ERK1/2、PI3K信号通路是否参与此分化过程。通过上述研究揭示TGF-β28608;活Myocardin家族诱导骨髓间充质干细胞分化及其分子机制,为临床应用和成体干细胞分化研究提供理论基础。

英文摘要: Bone mesenchymal stem cell (BMSC) is an adult stem cell that has multi-direction differentiation potential and is advantaged seed cell for tissue engineering. Myocardin-related transcription factors are novel coactivators for serum response factor (SRF). MRTF associating with SRF regulates smooth muscle cell special genes, which are the required cofactor during SMC development and differentiation. Based on our previous research, we will investigate if TGF-βtivates MRTF to induce BMSC differentiate into SMCs. The interaction site and the mechanism between MRTF and smad, which are signal transduction induced by TGF-βwill be addressed. The role of cyclin protein p16 and Rb will be researched. We will further study if p38, ERK1/2 and PI3K signal pathways are involved in this differentiation process. These researches will address the molecular mechanism of BMSC differentiation induced by MRTF and provide a theoretical basis for clinical application and adult stem cell differentiation research.
