中文摘要: 肠易激综合征(irritable bowel syndrome,IBS)是一种胃肠道运动、感觉及分泌异常的功能紊乱性疾病,其发病机制复杂。现已证实,IBS 患者体内多种生物活性物质异常及局部神经肌肉各种受体改变在IBS 发病中具有重要作用。本项目拟用全细胞膜片钳技术直接检测IBS 患者血浆对结肠平滑肌细胞动作电位、静息电位、钙依赖钾通道、延迟整流性钾通道、L-型钙通道 及5-羟色胺敏感钾通道电流的影响, 并应用放射免疫法检测IBS 患者空腹静脉抽取的血浆中5–T、MTL、CCK、SS、VIP、IL-6、CRH 及可能的生物活性物质含量的变化,同时探讨IBS 患者血浆中生物活性物质的紊乱导致肠动力异常的可能电生理机制,为寻找IBS 特异性治疗药物提供理论基础。

英文摘要: IBS(irritable bowel syndrome)is a kind of dysfunctional disease with complicated pathogenesis which involves abnormal intestine mobility, disturbance of internal organ perception and parasecretion of stomach intestine. It has been proved that the abnormality of many biological substances and the changing of the multiple regional neuro-muscle junction receptors play important role in the genesis of IBS.In this project, we will use the whole cell Patch Clamp to detect the influence of the plasma of IBS patient on the action potential, resting potential, and the current of calcium-dependent potassium channels, delayed rectifier potassium channels, L-type calcium channels and 5–T sensitive potassium channels of the colonic smooth muscle cells. We will also monitor the level of 5–T,MTL,CCK,SS,VIP,IL-6,CRH of the fasting vein blood from the IBS patients by using radioimmunoassay(RIA) method. And also, we intend to find out the possible electrophysiological mechanisms of the influence of the disorder of biological substances in the plasma of the IBS patients on abnormal intestine mobility, which may provide fundamental basis for finding the specific drug for IBS.
