中文摘要: 盾纤类纤毛虫是一类在海水养殖中造成重大危害的原生动物,目前尚无有效且安全的防治方法。形成休眠包囊是其抵御极端环境、传播及后期有效感染宿主的重要方式。对调控盾纤虫包囊形成与包囊解脱相关基因的研究是阻断其细胞分化关键基因的表达,抑制其生长、分化的前提。本研究拟通过对环境条件的控制实现对盾纤虫包囊形成与解脱的高效诱导,应用RNA-seq技术在转录组水平上研究盾纤虫营养期和休眠期基因表达的变化,以期寻找并克隆出影响休眠包囊形成的关键基因,以RNAi技术诱导盾纤虫形成包囊的关键基因沉默,观察基因沉默后的表型变化,注释相关基因的功能,了解其相互作用的方式,力图阐明盾纤虫营养期与休眠期细胞转化的机理,及应用RNAi方法对盾纤虫的关键基因表达进行干扰,从而达到抑制盾纤虫的生长与细胞分化的目的,为水产动物纤毛虫病的防制策略提供新的思路。

英文摘要: Some scuticocilates are pathogenic protozoa which do great harm to the marine breeding and culture. They can cause the important diseases in industrial fish-breeding, and at present time people still have not effective and safe method to defend and treat them. Their forming resting cyst is an important method to resist the extreme environment, spread, and effectively infect the hosts. It is a premise to study the key cell differentiation genes in vegetative cell and resting cyst of scuticocilates, and then we could try to disrupt their expression and afterwards suppress their growth and differentiation. This research intends to effectively induce scuticocilates to encyst and excyst depending on the control of environmental conditions. After that, in order to find out the key genes that influence the encystment and excystment, we plan to use RNA-seq to study the differential expression genes in vegetative cell and resting cyst on the level of transcriptome. Meanwhile, this study will use RNA interference technique to induce the key genes in cyst forming silent and observe the phenotypic changes after gene silence.
