中文摘要: 新基因从头合成起源一直被认为是很少发生的,但近期却引起人们的关注与重视。我们前期工作挖掘到60个人类特异的从头合成起源蛋白编码新基因,它们很多在大脑皮层与睾丸中表达很高,很可能已经与其它基因发生相互作用,形成新的网络途径,影响人类一些特异的适应性性状。本课题拟从中挑选在大脑皮层中表达较高的6个新基因,研究它们的生物功能和调控网络,分析它们是否参与重要的大脑发育调控途径。挖掘小鼠与黑腹果蝇基因组中的从头合成起源新基因,评估从头合成起源的普遍程度、产生速率。 探索如何利用从头合成起源新基因研究人类适应性进化的机制,多基因相互作用形成机制,以及基因网络的起源。

英文摘要: De novo origination of new genes is traditionally considered to be a very rare process; however this is a process that is now attracting greater attention. We identified previously 60 human specific de novo-originated protein coding genes, many of which demonstrated high expression levels in the cerebral cortex and testes. It suggests that they may have interactions with other genes to generate new networks, which influence some adaptive traits unique to human. This project will choose 6 new genes with high expression levels in the cerebral cortex, and study their biological functions and regulatory networks to see whether they play roles in some pathways responsible for brain development. We will search for de novo-originated protein coding genes in the mouse and Drosophila melanogaster genomes, to study the rate and universality of de novo origination. The project tends to explore how to study the mechanism of human adaptive evolution, the origin of multi-gene interaction, and the origin of gene network based on the de novo-originated new genes.
