中文摘要: 本项目申请人已研究了Maillard反应修饰鲢鱼寡肽的反应机制,发现Maillard反应较大提高了其在体外检测体系的抗氧化活性;并分离制备了水溶性高分子活性组分,初步分析了其结构特征。然而,Maillard反应虽能提高鲢鱼寡肽的抗氧化活性,也形成一些潜在的危害物质。针对Maillard反应修饰的鲢鱼寡肽在细胞和动物体内的构效关系及其潜在危害等问题,本项目拟通过Maillard反应制备水溶性高分子组分,深入定量定性分析Maillard反应形成的特征性产物,基于抗氧化活性、细胞毒性以及风险(risk/benefit)评价,实现其定向制备。然后,在抗氧化跟踪下,对其分离纯化并表征其结构,分析其对细胞氧化应激的保护及调节控、以及动物体内的抗氧化作用,揭示其抗氧化活性的构效关系。本项目的研究不仅能够推动关于肽类Maillard反应的理论的发展,还将为具有抗氧化功能的新型肽类保健食品开发提供指导。

英文摘要: In the present research, we have found that the Maillard reaction products from silver carp peptide and glucose showed higher antioxidant activity determined by free radical scavenging activity and the inhibition effects of lipid oxidation, and the water-soluble fraction in Maillard reaction products with higher antioxidant activity was obtained, and its structure characterization about the specific site between sugar and silver carp peptide during Maillard reaction was identified. However, Maillard reaction products possibly produced some potential harmful substances. Therefore, the research in the future project will focus on the structure of Maillard reaction products and their antioxidant activity in cell culture system and vivo (rats), and the potential cytotoxic effects. Firstly, the products formed at different stages of Maillard reaction will be identified and analysed in order to control the progress of Maillard reaction. Secondly, to analyse the water-soluble fractions of Maillard reaction products on cell viability, the cytotoxic effects and the evaluation of risk/benefit of Maillard reaction products based on its antioxidant activity and the contents of some potential harmful substances, and then prepare the safe water-soluble fraction of Maillard reaction products with higher antioxidant.
