中文摘要: 甜瓜是具有重要经济价值的葫芦科作物,在我国栽培广泛。由于我国东部广大季风区洪涝灾害频繁发生,甜瓜生产经常遭受严重危害,迫切需要培育耐涝优良甜瓜品种以满足生产需求。开展甜瓜耐涝性机理研究对甜瓜耐涝优良新品种的培育具有重要意义。课题组在前期研究中发现甜瓜种内存在耐涝性变异,并筛选出耐涝性差异较大的M19(耐涝型)和M63(涝敏感型)两份甜瓜材料。本研究计划利用M19和M63幼苗为主要研究材料,在人工淹水条件下,从解剖结构、生理生化特征等层次分析耐涝型甜瓜和涝敏感型甜瓜应对淹水胁迫的生理响应差异,采用表达谱测序技术(RNA-seq)从转录水平研究耐涝型甜瓜和涝敏感型甜瓜应对淹水胁迫的分子响应差异,筛选耐涝相关候选基因,并初步鉴定部分候选基因的功能。本项目的实施将有助于加深对甜瓜耐涝生理和分子机理的理解,丰富甜瓜耐涝相关理论,为挖掘甜瓜耐涝相关基因和培育耐涝优良品种奠定一定的理论基础。

英文摘要: Melon (Cucumis melon L.) is one of the most economically important and widely cultivated crops in China. In East China monsoon region, floods occurs frequently and results in reduction in melon production. Knowledge about the physiological and molecular mechanism of waterlogging tolerance is useful for melon breeding. Here two inbred lines, M19(waterlogging tolerant)and M63 (waterlogging susceptible), were selected for this study. Changes on anatomical, physiological, biochemical and transcriptional levels under waterlogging in melon will be investigated. Genes associated with waterlogging tolerance may be found and cloned. Functional identification of several genes will also be proceeded. The implement of the project will be helpful for elaborating the physiological and molecular mechanism of waterlogging tolerance in melon and be beneficial for melon breeding.
