中文摘要: 血小板的活化及其标记物的测定在生物材料的使用和心血管疾病的检测、预防、治疗以及与病理学研究等方面具有举足轻重的意义。本项目旨在结合近红外荧光成像、荧光共振能量转移和介入导管等技术,建立一套活体内在线研究介入材料表面与血小板相互作用的系统。首先选择活化血小板表面的P选择素(CD62P)和糖蛋白GP IIb/IIIa等标记物为检测目标,依据荧光共振能量转移的原理设计新型的在线荧光检测方法。然后选择医用聚氨酯为介入导管原料,通过表面形貌、官能团和仿生大分子等方面的设计和修饰,实时研究材料表面对血小板活化的影响。研究结果有望揭示血小板活化在血栓过程中的作用途径和机制,为介入材料的表面设计和血液相容性的改善提出新的思路,同时也为心血管疾病的预防和治疗开创新的途径。

英文摘要: The activation of platelets and detection of specific markers play important roles in evaluating the application of biomaterials. They are also significant in detection, treatment and prevention of pathological change in cardiovascular system. This project focuses on developing a near-infrared (NIR) fluorescence imaging catheter system which can explore the interaction between intervention materials and platelets in vivo. The system is composed of fluorescence imaging, fluorescence resonance energy transfer (FRET)-based sensor and interventional catheter. The platelet active labels such as P-selectin (CD62P) and GPIIb/IIIa were chosen as the targets monitored by the real-time optical immunosensor based on fluorescence resonance energy transfer. The key factors of medical PU catheter, such as topography, surface functional group and biomimetic interface, and their effects on platelet activation were investigated by the real time method. The results would reveal the pathways and mechanism of the platelet activation during the whole process when the thrombus is formed. It may offer new sights in designing biomaterial surface and improving blood compatibility of interventional materials. It would develop a novel method in the prevention and therapy of cardiovascular disease.
