中文摘要: 龟甲胶、鹿角胶是我国特有的常用传统动物药。目前商品龟甲胶、鹿角胶掺伪、造假情况多见,严重影响了该类药品的安全性和有效性。和大多数动物药一样,长期以来,由于其化学成分特殊性及质量控制技术的缺乏,尚无龟甲胶、鹿角胶的专属性检测方法,难以进行质量评价。申请者在前期研究中进行了阿胶特征肽的研究,形成阿胶的专属性检测方法。本课题拟采用蛋白酶切技术对龟甲胶、鹿角胶及常见的杂皮胶进行酶切,以UPLC-Q/TOF/MS进行肽图分析,以数学统计学对所有胶类LC-MS数据进行处理,找出龟甲胶、鹿角胶区别于其它胶类的特征肽;在此基础上,根据特征肽二级质谱进行测序并化学合成;建立以单体特征肽为对照的龟甲胶、鹿角胶专属性检测方法;同时开展与龟甲胶、龟角胶临床疗效相关的药理学研究,以期找到发挥药效作用的特征肽;针对筛选出的活性多肽,建立专属性的定性、定量方法,为该类药材的质量评价提供科学依据。

英文摘要: Tortoise shell glue and deerhorn glue are widely used as Traditional Chinese Medicines (TCM) in China, Adulteration and counterfeit of tortoise shell glue and deerhorn glue occured frequently,which affect the safety and effectiveness of these drugs. There are no specific method for idenfification of tortoise shell glue and deerhorn glue currently,because of the specificity of the constituents of drugs from animals and lack of the technologies to protein analysis, In previous study, we identified donkey-hide gelatin, bovine-hide gelatin, and pig-hide gelatin by principal component analysis, and we found marker peptides of donkey-hide gelatin, bovine-hide gelatin, which were amino acid sequenced and synthetized. The specific method for identification of donkey-hide gelatin was established. In our study, we will analyze the trypsin digested samples by UPLC-Q/TOF/MS, The UPLC-MS data of the trypsin digested samples will be subjected to principal component analysis (PCA) and an orthogonal partial least squares data analysis (OPLS-DA) to find marker peptide of tortoise shell glue, deerhorn glue. We will give the amino acid sequence of marker peptides base on the mass spectrometry of marker peptides and synthetise the marker peptides.
