中文摘要: 腈水解酶是一类高效的生物催化剂,可以将腈一步水解成羧酸,在生物制药和腈污染物降解过程中具有重要的应用。腈水解酶序列、结构与功能关系及在此基础上的腈水解酶的筛选和分子改造,迄今国内外尚无报导。本项目以腈水解酶为研究对象,系统研究腈水解酶序列、结构与功能的关系,建立为不同腈化合物的生物催化和转化需求筛选、设计不同的腈水解酶的方法。项目将在揭示序列、结构与功能的关系和腈水解酶与不同底物分子识别机理的基础上,发展和完善腈化合物生物催化和转化过程中生物催化剂发现的方法学;为开发新的重组腈水解酶或从头设计腈水解酶和对腈水解酶的分子改造方面提供新的技术方法和理论。

英文摘要: Nitrilase, as one of the important industrial enzymes in pharmaceutical and nitriles degradation, is a valuable biocatalyst hydrolyzing non-peptide carbon-nitrogen bonds in a single step. There is no any reports about the sequences, structures and functions relationship of nitrilase and its molecular modification studies. In this study, the sequences, structures and functions relationship of nitrilase and its molecular modification were investigated in detailed, to generate a new method for screening or redesign novel nitrilases. The new methodology in finding novel nitrilases will be constructed based on the mechanism of recognition between nitrilases and substrates, and relationship of sequences, structures and functions. This study will provide new methods and technologieson exploring and de novo redesigning novel recombinant nitrilases and molecular modification of nitrilases, which will pave foundation on fundamental research of nitrilases.
