中文摘要: 中药黄芪及其蜜炙品临床应用广泛,而其炮制机理尚未阐明。申请者根据前期建立了黄芪药材的指纹图谱分析方法,初步比较蜜炙黄芪炮制前后化学成分变化,发现8种炮制前后变化较大的化合物与其药效活性相关。本项目拟在此基础上应用UPLC/Q-TOF-MS技术和R语言的化学计量学方法,进一步研究蜜炙黄芪炮制前后有效成分作用于脾气虚动物模型的吸收、代谢情况,通过建立炮制前后引起体内变化的代谢物和免疫相关生化指标的关系来探索其药效作用机制,为阐明蜜炙黄芪的炮制机理及临床合理用药提供实验依据。

英文摘要: Astragalus membranaceus and its fried product with honey are the traditional Chinese medicines that have extensive clinical application, but their processing mechanism has not been elucidated. Based on the developed fingerprint method of Astragalus membranaceus, applicant preliminary compared the change of chemical composintion of Astragalus membranaceus and Astragalus fried with honey and found 8 differential compounds after processing. The project is planned using UPLC/Q-TOF-MS technology, combined with chemometrics methods of R language to research the absorption and metabolism of active constituent in Astragalus membranaceus and its fried product with honey. Pharmacodynamic mechanism affecting on spleen qi deficiency will be explored with the combination of biochemical indicators and changed metabolites in vivo. It has important significance to clarify the processing mechanism of astragalus.
