中文摘要: 果胶因其独特的化学结构具有天然的细胞识别活性,在生物医学应用中有望弥补壳聚糖和海藻酸盐的不足。赋予果胶纳米纤维的结构形态更利于提升其生物医用的效果与范围。本项目旨在发展具有天然的细胞识别活性的果胶电纺纳米纤维。通过以少量的柔性合成高分子聚氧化乙烯作为可移除的载体,解决果胶水溶液中高分子链间缠结不足的问题,实现果胶纳米纤维的电纺制备;通过对非活性的主链进行选择性交联,同时保持活性侧链自由可用,实现保持生物活性的前提下的果胶纳米纤维交联;进一步研究其理化和生物学性质,并探索其生物医学应用。本项目一方面将发展电纺制备果胶纳米纤维的方法和在保持生物活性的前提下交联果胶纳米纤维的方法,另一方面将开发一类能够耐受水相环境且具有天然的细胞识别活性的果胶纳米纤维材料,为天然高分子的生物医学应用提供材料基础。

英文摘要: Pectin exhibits intrinsic cell recognition activity due to its unique chemical structure and is promising for remedying the defects of chitosan and alginate for biomedical applications. Endowing pectin nanofibrous structure feature will further improve the efficacy and scope of its biomedical applications. In this project, we aim to develop electrospun nanofibers of pectin with intrinsic cell recognition activity. We will use a low content of flexible-chain synthetic polymer, poly(ethylene oxide), as removable carrier to overcome the problem of insufficient inter-chain entanglements in aqueous pectin solution and enable the fabrication of pectin nanofibers through electrospinning. We will crosslink the non-active main chains of pectin and retain its active side chains accessible to realize the crosslinking of pectin nanofibers without sacrificing their bioactivity. We will further examine the physiochemical and biological properties of pectin nanofibers and explore their biomedical applications. The project will on one hand develop approaches to fabricate pectin nanofibers through electrospinning and approaches to crosslink pectin nanofibers without sacrificing their bioactivity, and on the other hand develop a class of pectin nanofibers for biomedicine, which are capable of tolerating aqueous physiological environment and are of intrinsic cell recognition activity.
