中文摘要: 花的颜色是观赏植物的重要特征,花瓣基部的色斑是牡丹的重要特色之一,除了极具观赏价值外,色斑已成为牡丹品种群分类和牡丹进化研究的重要依据,但对其相关研究甚少。关于牡丹斑色形成的分子机理至今尚不清楚,亟待开展相关研究。本项目组前期对35个西北牡丹品种花瓣斑与非斑部分色素组成和含量等进行了测定和分析,从化学角度揭示了牡丹斑色的成因。因此,本项目拟在前期研究的基础上,试图利用高通量测序技术获得牡丹斑与非斑着色过程中的转录谱;分析差异基因表达谱,并对候选基因通过荧光定量PCR技术进行表达分析;通过病毒介导基因沉默技术(VIGS)从反向遗传学角度快速研究基因功能;结合植物化学和分子生物学研究结果,揭示牡丹斑色形成的化学和分子生物学机理。为牡丹斑色育种奠定基础,为其他被子植物斑色研究提供思路和依据。

英文摘要: Flower color is an important trait for ornamental plants. Blotches with different morphology and colors at the base of petal is one of the most important characteristics of tree peony, which has been the criteria for cultivar classification and evolution study besides its ornamental value. However, few research on the blotch color formation can be referred now. The molecular mechanism on the coloration of tree peony petal blotch is still unknown. We had analyzed the constituent and content of flower pigment of petals between blotch and non-blotch part from 35 cultivars belonging to XiBei cultivar group in our previous study, and revealed the coloration causes of petal blotch from the phytochemical point. Based on this study, we will attempt to obtain the transcript profiling from blotch and non-blotch part of tree peony petals through next-generation sequencing technique, and try to find the differential expression profile of genes which will be validated by qRT-PCR. Further, Virus-induced gene silencing technique will be used to characterize gene function in a rapid way from the perspective of reverse genetics. Finally, we will demonstrate the petal blotch coloration mechanism by combining the results of photochemistry and molecular biology, which will lay the foundation for the breeding of tree peony cultivars with novel petal blotch colors, and provide ideas and basis for the blotch color study on other angiosperm plants.
