中文摘要: 昼夜节律异常是抑郁症临床表现的重要方面。但是,目前抑郁症昼夜节律异常的脑功能影像学研究非常少,尤其缺乏在脑网络水平的发现。结合文献调研和前期研究,我们提出使用功能磁共振影像研究抑郁症脑功能昼夜节律异常。这对理解抑郁症的病理机制具有重要意义。申请人在前一个基金委青年基金的资助下研究了健康成年被试在昼夜不同时刻褪黑素浓度变化与静息态脑功能网络活动模式变化的关系。本项目中,我们将继续这一方向,并扩展研究抑郁症脑功能昼夜节律异常。具体来说,检测抑郁症病人组和正常对照组在1天3个不同时刻(7:00,15:00和23:00)血清激素浓度、静息态脑功能网络以及任务态脑功能网络的活动,然后以褪黑激素浓度作为人体内昼夜节律的生物标记,研究其浓度变化与静息态脑功能网络和任务态脑功能网络变化的关系,比较抑郁症病人组和对照组的脑功能网络活动模式昼夜变化的异同,发现抑郁症脑功能昼夜节律异常的脑网络表征。

英文摘要: Abnormal circadian rhythm is one of important aspects of the clinical manifestations of major depressive disorders (MDD). However, it is unclear of the relationship between the MDD and the diurnal variations in brain functional networks, which would be helpful for understanding the pathogenesis of MDD. Sponsored by natural science foundation of China, we have demonstrated the oscillations of the resting state brain functional networks together along with the circadian rhythm in healthy adult. In this proposal, we will continue along this direction, and extend it to exploring abnormal diurnal variations of brain function networks in the MDD. Specifically, we will measure some blood hormones and scan the resting/task state brain activity for both the MDD patients and the normal controls in three time points during one day (that is, 7:00, 15:00 and 23:00), and then utilize blood serum melatonin concentration as one internal biological marker of the circadian rhythm, and investigate the correlations between the changes of melatonin concentration and ones of the activity pattern of the brain functional networks in both the MDD patients and the normal controls, and finally characterize the abnormalities of diurnal variations of brain functional networks in the MDD patients.
